where and how do I specilfy the lot size?

For a live account, this is controlled through Gain. They will determine lot size at the account level and you submit orders in NinjaTrader with quantities 1, 2, 3, etc.

so, I do I distinguish between live account where mini lot size is 1 to simulated where minilot size is 10000?

For live trading, this is set at the account level. Some brokers will expect quantity just like this, where 100,000 is a standard unit. Other brokers may set where a quantity of 1 represents this. If you are unsure, please contact the broker you're working with, for the quantity expected when trading on your account.
